Activation-induced deaminase (AID) not only mutates DNA within the immunoglobulin loci to generate antibody diversity, but it also promotes development of B cell lymphomas. To tame this mutagen, we performed a quantitative high-throughput screen of over 90 000 compounds to see if AID activity could be mitigated. The enzymatic activity was assessed in biochemical assays to detect cytosine.

The activation-induced cytidine deaminase (AID) gene, specifically expressed in germinal center B cells in mice, is a member of the cytidine deaminase family. We herein report mutations in the human counterpart of AID in patients with the autosomal recessive form of hyper-IgM syndrome (HIGM2). Three If AID protein is found to be present and functional in these tissues (which remains to be demonstrated), the ability of AID to target non-Ig genes could have physiological roles in developmental processes and in evolution. It is possible that AID generates germline mutations and thereby contributes to genetic diversity and natural selection. Very recently, AID was implicated in programmed DNA. AID-generated somatic hypermutations affect the variable (V) regions of genes encoding immunoglobulins. Somatic (hyper)mutation affects only individual cells of the immune system, so the programmed mutations that it generates are transmitted only within the particular cell line (somatic) and are not transmitted to the organism's offspring

The cytidine deaminase AID hypermutates immunoglobulin genes but can also target oncogenes, leading to tumorigenesis. The extent of AID's promiscuity and its predilection for immunoglobulin genes. Immunology focuses on the human body's built-in defense system. In a healthy person, the immune system helps the body fight infection by rejecting foreign viruses and bacteria. When the immune system is defective, it can fail to protect the body, or even attack it. Diseases caused by disorders of the immune system may be caused by immunodeficiency, in which parts of the immune system fail to provide an adequate response, or autoimmunity, in which the immune system over.
